Snel opkomende SARS-CoV-2 B.1.1.7 sub-lijn in de Verenigde Staten van Amerika met spike eiwit D178H en membraan eiwit V70L mutaties
Samenvatting De SARS-CoV-2 B.1.1.7-afstamming is zeer besmettelijk en was vanaf april 2021 goed voor 92% van de COVID-19-gevallen in Europa en 59% van de COVID-19-gevallen in de VS. Het wordt gedefinieerd door de N501Y-mutatie in het receptorbindende domein (RBD) van het Spike (S)-eiwit en enkele andere mutaties. Deze omvatten twee mutaties in het N-terminale domein (NTD)van het S-eiwit, HV69-70del en Y144del (ook bekend als Y145del vanwege de aanwezigheid van tyrosine op beide posities). We hebben onlangs verschillende opkomende SARS-CoV-2-varianten van problemen geïdentificeerd, gekenmerkt door membraan (M) -eiwitmutaties, waaronder I82T en V70L. We identificeren nu een sublijn van B.1.1.7 die ontstond door opeenvolgende acquisities van M:V70L in november 2020, gevolgd door een nieuwe S:D178H-mutatie die voor het eerst werd waargenomen start februari 2021.
Het proportion B.1.1.7-isolaten in de VS dat tot deze sublijn behoort, is gestegen van 0,15% in februari 2021 tot 1,8% in april 2021. Tot op heden lijkt deze sublijn specifiek voor de VS te zijn met gemelde gevallen in 31 staten, waaronder Hawaï. Vanaf april 2021 vormde het 36,8% van alle B.1.1.7-isolaten in Washington. Fylogenetische analyse en transmissie-inferentie met Nextstrain suggereert dat deze sublijn waarschijnlijk afkomstig is uit Californië of Washington . Structurele analyse onthulde dat de S:D178H-mutatie zich in de NTD van het S-eiwit bevindt en dicht bij twee andere kenmerkende mutaties van B.1.1.7, HV69-70del en Y144del. Het is aan het oppervlak blootgesteld en kan de tertiaire configuratie of toegankelijkheid van NTD veranderen, en heeft dus het potentieel om neutralisatie door NTD-gerichte antilichamen te beïnvloeden.
Snelle driedimensionale vormbepaling van bolvormige eiwitten door capillaire elektroforese van mobiliteit en natieve massaspectrometrie
Gevestigde proteomics-methoden met hoge doorvoer bieden beperkte informatie over de stereostructuren van eiwitten. Traditionele technologieën voor het bepalen van de eiwitstructuur vereisen doorgaans moeizame stappen en kunnen niet op een high-throughput-manier worden uitgevoerd. Hier rapporteren we een nieuwe medium throughput-methode door mobiliteit capillaire elektroforese (MCE) en native massaspectrometrie (MS) te combineren voor de 3-dimensionale (3D) vormbepaling van bolvormige eiwitten in de vloeibare fase , die zowel de geometrische structuur als de moleculaire massa-informatie van eiwitten.
Er werd een theorie ontwikkeld om de hydrodynamische straal van ionen en de verdeling van de ladingstoestand in het natieve massaspectrum te correleren met geometrische eiwitparameters, waardoor een lage-resolutiestructuur (vorm) van het eiwit kon worden bepaald. Onze testgegevens van 11 verschillende globulaire eiwitten toonden aan dat deze benadering ons in staat stelt om de vormen van individuele eiwitten, eiwitcomplexen en eiwitten in een mengsel te bepalen en om veranderingen in eiwitconformatie te volgen. Naast het verstrekken van aanvullende informatie over de eiwitstructuur en het vermogen om mengsels te analyseren, is deze op MCE en native MS gebaseerde methode snel in snelheid en laag in monsterconsumptie, waardoor deze mogelijk toepasbaar is in top-down proteomics en structurele biologie voor intacte globulaire eiwit- of eiwitcomplexanalyse.

Snelle oppervlakte-immobilisatie van natieve eiwitten door katalysatorvrije amino-yn-klik-bioconjugatie
Oppervlakte-immobilisatie biedt een nuttig platform voor biosensing, screening van geneesmiddelen, weefselengineering en andere chemische en biologische toepassingen. Echter , sommige van de gebruikte reacties zijn inefficiënt en / of ingewikkeld, hun toepassingen te beperken in immobilisatie. Hierin gebruiken we een spontane en katalysatorvrije amino-yn-klik bioconjugatie om geactiveerde met ethynylgroep gefunctionaliseerde oppervlakken te genereren voor snelle immobilisatie van inheemse eiwitten en cellen.
Biomoleculen, zoals runderserumalbumine (BSA) , humaan IgG en een peptide van C(RGDfK), konden in slechts 30 minuten covalent op de oppervlakken worden geïmmobiliseerd. Met identify de biologische activiteit van de verankerde biomoleculen blijft intact, wat wordt geverifieerd door het efficiënt vastleggen van doelantilichamen en cellen uit de bulkoplossingen. Deze strategie vertegenwoordigt een alternatief voor zeer efficiënte biofunctionalisatie van het oppervlak.
Over de bindingsreactie van loratadine met humaan serum acute fase- eiwit alfa-1-zuur glyco- eiwit
Loratadine is een belangrijk anti-allergisch medicijn. Het is een antihistaminicum van de tweede generatie dat wordt gebruikt voor de behandeling van allergische rhinitis, hooikoorts en urticaria. Humaan alfa-1-zuur-glycoproteïne (AG) in serum is een belangrijk eiwit in de acute fase en de serumconcentratie ervan blijkt te stijgen bij ontsteking en acute respons. De bindingsinteractie tussen loratadine en AG wordt bestudeerd met behulp van spectroscopie en moleculaire koppeltechnieken. De resultaten verkregen uit fluorescentie- uitdovingsexperimenten toonden aan dat de fluorescentie-intensiteit van AG wordt gedoofd door loratadine. Loratadine bleek AG binden aan de bindingsconstante van ≈10 4 bij 298 Okay.
De vrije energieverandering van Gibb bleek negatief te zijn voor de interactie van loratadine met AG, wat aangeeft dat het bindingsproces spontaan is. Binding van loratadine met AG induceerde geordende structuren in het eiwit. Waterstofbinding en hydrofobe interacties waren de belangrijkste bindingskrachten tussen AG-loratadine, zoals blijkt uit moleculaire koppelingsresultaten. Deze studie suggereert het belang van het ruimtelijk binden van anti-allergische geneesmiddelen aan AG bij ziekten waarbij de plasmaconcentratie van AG vele malen toeneemt en interactie met dit eiwit vital wordt. Gerichte Protein Degradatie door middel van Quick optogenetische Activering en de toepassing ervan op de beheersing van de Cell Signaling
De ontwikkeling van methodologieën voor optisch getriggerde eiwitafbraak maakt de studie mogelijk van dynamische eiwitfuncties, zoals die welke betrokken zijn bij celsignalering , die moeilijk te onderzoeken zijn met traditionele genetische technieken. Hier beschrijven we het ontwerp en de implementatie van een nieuw lichtgecontroleerd peptide-degron dat N-end-pathway-degradatie aan zijn eiwitdoelwit verleent. De degron omvat een fotogekooid N-terminaal aminozuur en een lysinerijke linker met 13 residu’s. Door het N-terminale residu te kooien , waren we in staat om de herkenning van N-degron door een E3-ligase optisch te regelen, waardoor de ubiquitinatie en proteasomale afbraak van het doeleiwit werd gecontroleerd.
We demonstreren brede toepasbaarheid door deze benadering toe te passen op een various reeks doeleiwitten, waaronder EGFP, vuurvliegluciferase, de kinase MEK1 en de fosfatase DUSP6 (ook bekend als MKP3). De gekooide degron kan worden gebruikt met minimale eiwittechnologie en biedt vrijwel volledige, door licht geactiveerde eiwitafbraak op een tijdschaal van seconden tot minuut.
